What steps can I take if I do not have a sufficient amount of total RNA? (For service users)

How do I optimize my pre-hybridization? (For end users)

Spot says: Slide preparation is essential for eliminating high background signal. Here are a few tips for effective pre-hybridization of OneArray microarrays:

1. Recent changes in the pre-hybridization protocol can improve spot homogeneity and reduce high background signal. Our most recent modification incorporates an ethanol pre-wash that greatly improves the quality of data. Always check for the latest protocol on our Web site Support page.

2. Excessive jarring or mishandling of the microarrays can lead to aberrant spot morphology. This is often times the result of impact or extreme shear forces at the surface of the microarray. The ethanol pre-wash (see above) acts to eliminate some of these susceptibilities; careful handling is always recommended.

3. Unlike many other protocols, our pre-hybridization solution does not contain formamide. PhalanxBio scientists have found formamide can contribute to high background signal if included in the pre-hybridization mixture. (Howerver, keep in mind that fresh, de-ionized formamide IS included in the hybridization solution.)

4. A good source of BSA is essential for efficient slide preparation. Use at least molecular biology grade BSA (for example, electrophoresis grade is not effective). In addition, check to ensure you are using 1% BSA for pre-hybridization; this corresponds to 10mg/mL.

Where can I get the array files for an Agilent® scanner? (For end users)

Spot says: I have prepared .gal files compatible with Agilent® scanners in addition to the existing files compatible with GenePix™ software. Email spot@phalanxbiotech.com.
